As an agent of ChromaCon, Beijing huideyi has established Beijing laboratory & training center in Beijing changping life science park in 2017, and has successfully held several continuous flow technology training courses. In response to the requirements of customers in south China, huide yi will hold the autumn continuous flow technology training course in the biological incubator of shenzhen high-tech zone from September 19 to 20, 2018.
By then, huideyi will also bring Contichrom Discovery chromatography system, QuikSep EC manual chromatography column, QuikSep SC simple chromatography system and other sets of equipment to appear together, which can provide operation demonstration for the majority of customers.

? ? ? Capture SMB是將第一根層析柱的抗體/蛋白流穿繼續上樣至第二根層析柱，待第一根層析柱完全滿(mǎn)載后，方結束其上樣進(jìn)行淋洗，并繼續進(jìn)行洗脫、再生、再平衡操作，而此時(shí)第二根層析柱會(huì )獨立進(jìn)行上樣操作。通過(guò)切換柱位操作，保證2根層析柱的洗脫、再生、上樣等操作交替銜接進(jìn)行，從而實(shí)現工藝與系統硬件設計的完美結合，使填料利用率最大化，累積用量大為縮減。

MCSGP（多柱逆流溶劑梯度純化）
? ? ??MCSGP（Multicolumn Countercurrent Solvent GradientPurification）用于精純環(huán)節，可有效克服SMB（模擬移動(dòng)床）僅適用于二元分離、無(wú)法實(shí)現線(xiàn)性梯度的缺點(diǎn)。
? ? ? ?傳統的層析精純工藝，必須在收率和純度之間做出權衡，兩者此消彼長(cháng)。如何在保證獲得高純度產(chǎn)品的同時(shí)，得到高收率，一直是層析精純工藝所面臨的難點(diǎn)。
? ??? MCSGP工藝，可將含有不純組分的產(chǎn)品回收在一個(gè)封閉的環(huán)內，進(jìn)行循環(huán)式的層析操作，從而最大限度地提取純產(chǎn)品，使得收率和純度二者兼得。
應用案例
? ??? ?N-Rich工藝是微量物質(zhì)的富集過(guò)程，比如將產(chǎn)品的相關(guān)雜質(zhì)選擇性地富集在一個(gè)循環(huán)的層析過(guò)程中，待富集至目標量，再進(jìn)行洗脫回收。該方法特別適用于分離與產(chǎn)品有關(guān)的雜質(zhì)，如異構體和聚集體，并同時(shí)用于發(fā)現研究。

Discovery技術(shù)簡(jiǎn)介
? ? ? Discovery層析技術(shù)可將多步層析整合為一步，從而縮短純化時(shí)間，提高純化效率。第1步層析的產(chǎn)物無(wú)需進(jìn)行收集，通過(guò)在線(xiàn)稀釋?zhuān)芍苯由蠘又恋?根層析柱。即第1步層析的洗脫與第2步層析的上樣同時(shí)進(jìn)行，使純化周期顯著(zhù)縮短。ContichromDiscovery系統可額外擴展出35個(gè)入口，實(shí)現高通量純化。
應用案例
? ? ? 第一步Protein A；第二步CIEX；第三步Desalting（分子篩脫鹽換液）。
? ? ? ?在該過(guò)程中，Protein A的洗脫組分經(jīng)過(guò)在線(xiàn)稀釋?zhuān)苯由蠘又罜IEX層析柱，CIEX進(jìn)行線(xiàn)性洗脫，去除降解物及聚集體，將目標組分直接上樣至脫鹽柱。三步層析緊密銜接，前一步層析的洗脫與后一步層析的上樣同時(shí)進(jìn)行，使得純化效率顯著(zhù)提高。
? ? ? ?最終產(chǎn)品經(jīng)SE-HPLC檢測，純度高達99.5%，二聚體含量?jì)H為0.5%，比Protein A - Desalting兩步層析所得產(chǎn)品的二聚體含量降低近6倍，該產(chǎn)品可直接用于氨基酸序列測定、二硫鍵測定以及糖型分析等高純度要求的檢測。
? ? ? ?Discovery高通量（HT）型系統，采用Protein A - CIEX- Desalting三步向導模塊，每次上樣10 mL單抗上清液（1.0 mg/mL），每次產(chǎn)量均≥6.5 mg。單個(gè)樣品的三步純化耗時(shí)約為74 min，在24 h內純化出多達19個(gè)樣品，而且該時(shí)間包含了層析柱的清洗環(huán)節。該系統持續運行5天，純化樣品總數量高達91個(gè)。
